Using an expanded genetic code, the human lipocalin 2 (Lcn2) was functionalized with the unnatural amino acid boronophenylalanine and analyzed spectroscopically and structurally with regard to the binding properties of sugars. Furthermore, an attempt was made to select an aminoacyl-tRNA synthetase from synthetic gene libraries for the selective recognition of 7-hydroxycoumarylalanine. In addition, the Release factor 1 in the genomes of E. coli MG1655 and BL21 was edited to increase the efficiency of unnatural amino acid incorportation.
