I investigated the virus-host interaction of SARS-CoV-1/2, MPXV and IAV by integrative analyses of virus interactome, and virus-induced changes in mRNA and protein abundance, protein phosphorylation, ubiquitination, or turnover. I revealed key virus-perturbed pathways, e.g. the TGF-β signalling by SARS-CoV-2 and MPXV, and identified innate immunity modulators, e.g. the splicing factor GPKOW. I further tested several host-directed antivirals and the mRNA-based IFNλ1 therapy in vitro and in vivo. Altogether, my work improves our understanding of SARS-CoV-1/2, MPXV and IAV and enhances our pandemic preparedness.
