The influence of the cell adhesion molecule L1 (L1CAM) on the metastatic potenital of tumor cells was analyzed in an experimental xenograft metastasis model by knock down of endogenous tumor-cell expressed L1CAM, using shRNAi Technology. To elucidate differences in the contribution of
full-length L1 and its splice variant to metastasis, either isoform was selectively over-expressed in tumor cell lines of different origin. In contrast to its splice variant, over-expression of the
full-length isoform enhanced the metastatic potential of two tumor cell lines of different origin and suppression of both isoforms in an ovarian cancer cell line reduced tumor cell invasiveness in vitro and in vivo.
