The thesis in hand focuses on uPA and PAI-1 determination in small amounts of breast cancer tumor tissue and comprises a methodical and a clinical part. Methodically, we developed a new micro determination procedure (micro ELISA). We were able to depict that uPA and PAI-1 can be determined from five to ten 90µm cryostat sections of one frozen breast cancer tissue block or from core biopsies using commercially available and standardized ELISAs (American Diagnostica, Stamford, CT, USA). This miniaturized ELISA technique allows rapid and reproducible uPA and PAI-1 determination even in small tumor specimens or preoperative core biopsies. Clinically, we were able to demonstrate that PAI-1, but not uPA, in surgical specimens after primary systemic therapy may function as a surrogate marker to clinical response.
