Coxsackievirus B3 (CVB3) is one of the most common agents associated with viral heart disease. Viral proteases, important for viral maturation, are able to induce cytopathic effects by interaction with host specific proteins. This dissertation provides evidence for an interaction of CVB3 with the IL-6-type-signaling-pathway. Cleavages of IL-6-signaling pathway elements (STAT3 and gp130) are demonstrated by westernblots and immunofluorescence, following coxsackieviral infection. As a result phosphorylated products like pSTAT3Tyr705 and pSTAT3Ser727 are diminished as well as the IL-6 dependend expression of cardioprotective genes of the Bcl2-family (Mcl-1, Bcl-xL). Since IL-6 signaling is crucial for cellular immune response and survival, it displays an attractive aim of coxsackieviral proteases regarding infection, propagation and persistence.
