The transcriptional regulation by antitermination is an important control mechanism for the biosynthesis of ribosomal RNA in Escherichia coli and most other eubacteria. The interaction of NusB resp. NusB/NusE protein with boxA RNA, which is a key element of the antitermination complex of Escherichia coli, was studied by nuclear magnetic resonance spectroscopy and surface plasmon resonance. These in vitro studies led to first insights in respect of structural components involved in the diverse protein-protein and protein-RNA interactions, respectively. Besides the NusB of Escherichia coli the homolog protein of Thermotoga maritima was studied by crystallisation and X-ray diffraction measurements.
