The heat shock protein Hsp90 plays a key role in protein homeostasis in the cell. With the help of its numerous cochaperones it processes key players of cellular signaling cascades. In this work, the subgroup of TPR cochaperones and their role in the Hsp90 cycle was analyzed. Using a bioinformatic search algorithm new TPR cofactors were found and characterized in the model organism Caenorhabditis elegans and in humans. The mechanism which regulates the TPR cofactor protein phosphatase 5 in complex with Hsp90 was studied thoroughly. Using a high-throughput screening, five specific activators of this enzyme were identified and their mode of action revealed on molecular level. In addition, further insight of the TPR cochaperone UNC-45 in muscle maintenance was obtained.
