Ausgehend von der Beobachtung, dass die kutane Besiedlung mit
Staphylococcus aureus von Bedeutung für entzündliche und immunologische Reaktionen beim atopischen Ekzem ist, wurde untersucht, ob von den Bakterien gebildetes Protein A eine Aktivierung der Tumor- Nekrose- Faktor-alpha (TNFα) - Signalkette initiieren und/oder unterhalten kann. Es gelang der Nachweis des in der Keratinozyten- Membran lokalisierten TNFα- R1 Rezeptors. Die anschließenden Versuchreihen wurden an der standardisierbaren HaCaT- Zelllinie durchgeführt; es kamen immunbiochemische Techniken (Western Blot, Immunpräzipitation, RT- PCR, ELISA) und immunhistochemische Verfahren zur Anwendung. Hiermit konnte nachgewiesen werden, dass Protein A an den TNFα- R1 bindet und wichtige Mitglieder der TNFα- Signalkette wie JNK, MEK, p 38, NF-κB und AP-1 aktiviert. Nach Aktivierung dieser Signaltransduktion wird u.a. vermehrt Interleukin- 8 (IL- 8) und Cyclooxygenase-2 (COX-2) produziert; diese für Entzündungsreaktionen wichtigen Mediatoren ließen sich nach Inkubation von HaCaT- Keratinozyten mit Protein A mittels RT- PCR und ELISA nachweisen.
