Thermoplasma acidophilum were fractionated on Hydroxyapatite, MonoQ and native PA gels. Using the newly developed ‘Grid-blotting’ technique, EM samples were directly extracted from the focussed protein bands within the gel. Sample processing, from cell disruption until the final EM fixation step, was accomplished in less than 48 h. For future automation approaches of the ‘Grid-blotting’ technique, a gel processing platform was developed. This robot facilitates protein detection in PA gels based on the intrinsic fluorescence of proteins. Thus, the usage of tags or staining procedures is avoided.
