Cks1 ist ein Adapterprotein, das für die Bindung von Threonin187-phosphoryliertem p27Kip1 an den Ubiquitin-Ligase-Komplex SCFSkp2 essentiell ist. Im Rahmen dieser Dissertation sollten Funktionen von Cks1 unabhängig vom Abbau von p27Kip1 identifiziert werden. Dafür wurden p27Kip1-/-; Cks1-/- Mäuse generiert und deren Phänotyp in vivo und in vitro untersucht und mit dem des p27Kip1-/- verglichen. Es konnte gezeigt werden, dass Cks1-Verlust unabhängig vom p27Kip1 Genotyp zu einer signifikant verminderten Proliferation führt. Cks1 ist, unabhägig von SCFSkp2, essentiell für den geregelten G1/S Übergang und wirkt p27Kip1-abhängig am G2/M Übergang. Die Tumorsuppressorfunktion von p27Kip1 wird durch den zusätzlichen Verlust von Cks1 nicht beeinträchtig.
