Antibodies are key molecules of the humoral immune response and of outstanding biotechnological interest. Within this work, novel insights into the folding and assembly process of IgG antibodies could be obtained. The atomic resolution description of the folding pathway of an isolated IgG domain revealed unexpected structural features of a folding intermediate which guide the folding reaction and render it robust. Studies on antibody fragments provided, for the first time, detailed insights into the molecular mechanisms of antibody quality control in the cell which will pave the path for rational optimization of antibody molecules.
