The inositol 1,4,5-triphosphate receptor-associated cyclic GMP (cGMP)-kinase substrate IRAG is part of the NO/cGMP-signalling mediating the relaxation of smooth muscle and thereby modulating vascular tone.
Calcium measurements with fura-2 in cultured aortic vascular smooth muscle cells (VSMCs) showed that Norepinephrine-induced increase of intracellular calcium concentration is suppressed by the membrane-permeable cGMP analogue 8-Br-cGMP in VSMCs of wild-type, but not of IRAG-knockout mice (IRAG-/-). Furthermore western blots indicated an altered protein setting in arterial cells of IRAG-/-.
These results account for the importance of IRAG for the cGMP-dependent calcium regulation in VSMCs. In addition, we established a new method for calcium measurements in murine freshly isolated VSMCs.
