This paper examines the role of the urokinase-type plasminogen activator uPA and its type 1 inhibitor PAI-1 in invasion and metastasis. In a primary breast carcinoma (n=73), uPA and PAI-1 were determined in the tumour and in a corresponding affected or not affected axillary lymph node using ELISA. Significantly higher uPA concentrations were observed in the primary tumour than in lymph nodes; higher PAI-1 in the lymph node was accompanied by a significantly worsened prognosis. These results support data on a key role of PAI-1 and uPA in the process of the formation of metastases and open new therapy options for breast carcinoma.
