Abstract:
The Human Adenovirus oncogene E4orf1 is necessary and sufficient to activate cellular Akt of the PI3K-Akt pathway in human lung epithelial cells and is required for viral progeny production without impacting other viral markers. Human p53 interacts and destabilizes E4orf1, negatively impacting Akt phosphorylation, which highlights p53’s antiviral function against E4orf1 mediated cellular proliferation or transformation. This study reveals that E4orf1 interacts with the adenoviral E3 ubiquitin ligase, modulating ubiquitin modification of targets like p53.

Das Onkogen E4orf1 Humaner Adenoviren ist nötig und ausreichend um zelluläres Akt des PI3K-Akt Wegs in Lungenepithelzellen zu aktivieren und bedingt die Produktion viraler Nachkommen, ohne andere virale Marker zu modulieren. p53 interagiert mit und destabilisiert E4orf1, was die Akt-Phosphorylierung hemmt. Dies suggeriert eine antivirale Funktion von p53 gegen E4orf1-vermittelter zelluläre Proliferation und Transformation. Die Studie zeigt, dass E4orf1 mit der viralen E3 Ubiquitin Ligase interagiert und die Ubiquitin-Modifikation von Zielproteinen wie p53 moduliert.
