Kurzfassung:
In dieser Arbeit wurde untersucht, welche Möglichkeiten bestehen, die Konzentrationen von Nε-Carboxymethyllysin (CML) im Organismus zu reduzieren. Einerseits konnte gezeigt werden, dass über therapeutische Verabreichung die CML-Konzentration im Organismus diabetischer Ratten verringert werden konnte. Andererseits wurde über Zellkulturstudien gezeigt, dass in Gegenwart von mit CML-angereichertem Casein in Caco-2-Zellen ein erhöhter endosomaler Abbau stattfand und die Behandlung der Zellen mit Casein, CML-angereichertem Casein und CML eine Hitzeschockantwort aktivierte. Da weiterhin eine Aufnahme von Casein, CML-angereichertem Casein und CML in die Zellen gezeigt werden konnte, ist ein Abbau dieser Proteine über diese Schutzmechanismen denkbar.

The aim of this work was to identify mechanisms that decrease Nε-Carboxymethyllysin (CML) contents in vivo. On the one hand it could be shown that therapeutic treatments of diabetic rats were effective. On the other hand cell culture studies revealed that in human intestinal Caco-2 cells, endosomal degradation mechanisms were activated in the presence of CML-fortified Casein. Beyond that, a heat shock answer was demonstrated after the treatment of the cells with Casein, CML enriched Casein or CML. In accordance with results indicating a cellular uptake of Casein, CML enriched Casein as well as CML by Caco-2 cells, a degradation of these proteins by the mechanisms of the proteasome system are conceivable.
