Abstract:
Amyloid β (Aβ) plays a central role in the pathogenesis of Alzheimer’s disease (AD). Aβ‘s excessive accumulation can be hindered by therapeutically blocking the β-secretase 1 (BACE1) enzyme involved in its production. In an AD mouse model, I combined two-photon and widefield calcium imaging to show that BACE1 inhibition repairs dysfunctional brain circuit activity. The chronic BACE1 treatment recovered neuronal hyperactivity, slow waves‘ coherence, and memory defects. This is encouraging concerning clinical primary prevention strategies being currently tested.

Amyloid β (Aβ) spielt eine zentrale Rolle in der Pathogenese der Alzheimer-Krankheit (AD). Die Hemmung der β-secretase1 (BACE1), welche an Aβ‘s Produktion beteiligt ist, gilt als attraktiver Therapieansatz. Ich habe die Zwei-Photonen-, Widefield-Bildgebung und Verhaltenstests in einem AD-Modell kombiniert, um den Effekt einer BACE1-Hemmung auf Funktionsstörungen in AD zu untersuchen. Die BACE1-Hemmung reparierte zelluläre und langreichweitige Netzwerke, sowie Gedächtnisstörungen. Dies ist ermutigend im Hinblick auf aktuell getestete klinische präventive Studien.
