Abstract:
Im Rahmen der vorliegenden Arbeit wurden verschiedene Methoden zur Immobilisierung von Antikörpern auf einer Goldelektrode untersucht, mit dem Ziel, einen amperometrisch auslesbaren Immunosensor zu entwickeln. Das Messsystem basiert auf Siliciumchips mit Goldinterdigitalelektroden im submikrometer Bereich und einem Multikanalpotentiostaten zur Auslesung des amperometrischen Sensorsignals im nA-Bereich. Dieses wird durch Redoxrecycling des aus p-Aminophenylphosphat enzymatisch generierten Mediatormoleküls p-Aminophenol erzeugt. Zur Immobilisierung der Antikörper wurden verschiedene Oberflächenbeschichtungen gewählt, auf denen der Antikörper nachfolgend chemisch oder physikalisch gebunden wurde. Hauptkriterien für die Bewertung der Beschichtungsmethode waren dabei die Signalintensität und die Differenzierbarkeit zwischen Positiv- und Negativsignal bei der nachfolgenden Messung. Zunächst wurde ein Cysteamin-Monolayer mit nachfolgender kovalenter Antikörper-Bindung getestet. Dieses System führte zu einer sehr definierten, dichten Oberflächenbeschichtung mit sehr guter Differenzierbarkeit bei jedoch recht geringer Signalintensität. Die Beschichtung mit Avidin und Biotin-gekoppeltem Antikörper erfüllte die Erwartungen in der Signaldifferenzierbarkeit nicht. Als Methode der Wahl erwies sich die Beschichtung mit Protein A, welches den Antikörper Fc-spezifisch an der Oberfläche fixiert und zu sehr hohen Signalintensitäten bei guter Differenzierbarkeit führte. Diese Erkenntnis wurde auf ein System von praktischer Relevanz übertragen und ein Testaufbau zum Nachweis von spezifischem IgE in menschlichem Serum entwickelt, welches im Falle einer allergischen Erkrankung im Blut zirkuliert. Erste Tests haben gezeigt, dass diese Methode als in vitro Allergietest angewendet werden kann.

The objective of this work was the investigation of different surface coating methods in order to immobilize an antibody layer on interdigital gold electrodes and establish the best suited test design which can be applied as an immunosensor. For the measurements a very sensitive amperometric biosensor based on interdigitated gold electrode arrays (IDA) made in silicon technology was used. The electrical signal read out of the immunoanalytical reaction (antigen antibody binding) is based on the redox recycling of the mediator molecule p-Aminophenole: An alkaline phosphatase-labeled detection antibody initiates the enzymatic liberation of p-Aminophenole from p-Aminophenylphosphate, the resulting current in the nA range is amplified and detected by a multichannel potentiostat. To immobilize antibodies on a gold surface a so-called interlayer has to be prepared on which the antibody was chemically or physically coupled. In order to get a high sensor signal and a good differentiation between positive and negative signals different approaches for the immobilization of the antibody layer on the gold electrode surface and their effects on the electrical read out have been investigated. The specific binding of the antibody via Cysteamine self-assembled monolayers (SAM) on the gold surface followed by a covalent peptide coupling led to a well defined surface. Alternatively a semi-specific binding of the antibody with avidin-biotin-technology was investigated. The gold electrode surface was coated with an unspecific adsorbed avidin-agarose-layer and then specifically coupled to a biotinylated antibody. The most promising test design for a specific antibody immobilization was found in the Protein A-antibody-system: The electrode surface was coated with a soluble Protein A which is known to bind strongly to the Fc-fragment of IgG. The specifity of the enzymatic read out with p-Aminophenylphosphate using Protein A for the immobilization of the catcher-antibody was high. In next steps the Protein A-antibody-system has been testet for the detection of specific IgE in Human serum for the application as in vitro allergy testing.
