E-cadherin ist ein Zelladhäsionsmolekül, das bei Magenkarzinompatienten häufig mutiert ist. Gegen die beiden Hauptmutationen, Deletion von Exon 8 bzw. Exon 9, sind monoklonale Antikörper verfügbar. In dieser Arbeit wurden funktionale, rekombinante, disulfidstabilisierte Immuntoxine mit Pseudomonas Exotoxin A als Effektormolekül generiert und auf Spezifität der Bindung und Zytotoxizität getestet. Es konnte gezeigt werden, dass die zwei E-cadherin Deletionsmutanten ausgezeichnete, tumorspezifische Zielmoleküle für eine mögliche Tumortherapie bei einer Subgruppe von Magenkarzinompatienten darstellen.

The cell adhesion molecule E-Cadherin is frequently mutated in stomach cancer patients. Against the two main mutations, deletion of exon 8 or exon 9, respectively, there are monoclonal antibodies available. In this work, functional recombinant disulfide-stabilized Immunotoxins with Pseudomonas Exotoxin A as effector molecule have been generated and analyzed for specificity of their binding and cytotoxicity. It could be demonstrated, that the two E-Cadherin deletion mutants are excellent, tumor specific target molecules for a potential tumor therapy for a subgroup of stomach cancer patients.
