Abstract:
Der Effekt des alternativen Spleißens auf die Struktur und Funktion der meisten Proteine ist unklar. In der Doktorarbeit wurden alternative Spleißvorgänge für die humanen Sirtuine und alpha-Kristalline identifiziert und auf Transkriptlevel nachgewiesen. Sirt6 Isoform 1 und 2 hatten ähnliche Sekundärstrukturen und konnten beide ein Substratpeptid binden. Dennoch war nur Isoform 1 in der Lage, dieses zu deacylieren. Die alternative Spleißvariante alpha A-Kristallin Isoform 2 hatte ebenfalls eine vergleichbare Sekundärstruktur zur kanonischen Isoform, bildete aber Monomere und Dimere. alpha A-Kristallin Isoform 2 modulierte die Oligomergröße und Chaperonaktivität der kanonischen alpha-Kristalline.

The effect of alternative splicing on the structure and function of most proteins is obscure. Here alternative splicing events for human sirtuins and alpha-crystallins were identified and verified on transcript level. Sirt6 isoform 1 and 2 had a similar secondary structure and both could bind a substrate peptide. However, only isoform 1 was able to deacylate the peptide. The alternative splice variant alpha A-crystallin isoform 2 also had a comparable secondary structure to the canonical isoform, but formed monomers and dimers. alpha A-Crystallin isoform 2 modulated the oligomeric size and chaperone activity of the canonical alpha-crystallins.
