The human Fic enzyme FICD regulates the activity of the chaperone BiP by reversible AMP transfer. Herein, I use thiol-reactive nucleotide derivatives (TReNDs) to covalently link FICD to its substrate BiP and show that the target specificity of FICD is mediated by its TPR motifs. Furthermore, I use TReNDs for the identification and characterization of novel FICD AMPylation substrates. Preliminary work on a concept that allows covalent enrichment of AMPylation substrates is presented as well as an overview on recent advances on the covalent stabilization of macromolecular complexes for structural biology.
