The aim of the thesis work was to access rare antigen specific T cells from a healthy donor repertoire. With a combination of existing and recent methods for enrichment and extraction of TCRs (T cell receptors), we were successful in establishing a workflow for multimer based T cell identification, screening and extraction of TCR sequences. In conclusion, using a more economic workflow we were able to identify and isolate a broad spectrum of TCRs for different tumor and viral antigens for future applications.      «

Das Ziel der Diplomarbeit war seltene Antigen-spezifische T-Zellen aus einem gesunden Spender Repertoire zuzugreifen. Mit einer Kombination aus bestehenden und kürzlich Verfahren zur Anreicherung und Gewinnung von TZR (T-Zell-Rezeptoren) waren wir erfolgreich in einen Workflow für Multimer basierte T-Zell-Identifizierung Festlegung, Screening und Extraktion von TCR-Sequenzen. Abschließend einen wirtschaftlicheren Workflow konnten wir ein breites Spektrum an TCRs für verschiedene Tumor und virale Antigene für zukünftige Anwendungen zu identifizieren und zu isolieren.     «
