The aim of the thesis work was to access rare antigen specific T cells from a healthy donor repertoire. With a combination of existing and recent methods for enrichment and extraction of TCRs (T cell receptors), we were successful in establishing a workflow for multimer based T cell identification, screening and extraction of TCR sequences. In conclusion, using a more economic workflow we were able to identify and isolate a broad spectrum of TCRs for different tumor and viral antigens for future applications.
