The influence of guanosinerich DNA oligonucleotides (ODN) on T cell activation in vitro was investigated in this study. When triggered via their T cellreceptor (TCR), guanosinerich ODN evoked IL-2 driven T cell proliferation. ODN induced T cell proliferation was strictly dependent on a TCR signal and mimicked costimulation via CD28. However, the same ODN showed inhibitory activtiy on cell growth in a dose dependent manner. This inhibitory activity was also pronounced in primary T cells and T cellines, but varied depending on the mitogenic stimulus. While guanosinerich ODN did not interfere with early TCR signalling events , a blockade of costimulatory signals other than the physiological costimulator CD28 seemed to be feasible.
