SUMO1, an ubiquitin-related protein, regulates important cellular processes via post-translational protein modification. In context with the substrates PCNA and IkBa SUMOylation is discussed as an antagonism for ubiquitination. Besides an extensive mutagenesis analysis of SUMO1, poly-ubiquitination of non-conjugated and conjugated SUMO1 is demonstrated for the first time. In addition rapid proteasomal degradation could be excluded for SUMO1. These surprising results implicate conjugated SUMO1 as an alternative ubiquitiniation-site and therefore potentially as a signal for degradation.
