The mechanism of the activation of the serin/threonine kinase Akt induced by the adapterprotein Grb10 involves the localization to the plasma membrane. Additionally, a Yeast-Two-Hybrid Screen revealed the interaction between Grb10 and 14-3-3. This interaction depends on phosphorylation and the binding site in Grb10 could be identified. It may be said that, Grb10 interacts with Akt in the cytosol. After receptor stimulation, the complex recruits to the plasma membrane. The binding of Grb10 and 14-3-3 regulates the activation of Akt.
