Interferon-gamma is a inflammatory cytokine with essential functions in innate immunity. By employing a method based on subtractive hybridisation, a murine macrophage cDNS library was generated, that contains differentially IFNg inducible genes. One unknown gene, PUMA-G (protein upregulated in macrophages by IFNg), that was preferentially inducible by IFNg in macrophages, was cloned and characterised. PUMA-G is a novel member of the G-protein-coupled receptor superfamily. For analysis of the in vivo function of PUMA-G, the gene was inactivated in the mouse by homologous recombination of embryonic stem cells.
