Protein structure determination by NMR spectroscopy is becoming more and more important in modern research. The assignment process is time consuming and contains many repetetive steps. Therefore, it is suitable for automatization. In this thesis methods for the automated assignment of heteronuclear NMR spectra of proteins have been developed and tested with two proteins. Two software packages, PASTA V3.0 (Protein Assignment by Threshold Accepting) and PASTA Toolkit, have been realized and applied during the assignment of VAT-N (183 amino acids). A new methodology for the assignment of protein complexes using data from a structurally homologue complex of the same protein has been outlined and tested during the backbone assignment of the NADPH-complex of DHFR from E. coli.
