In this dissertation the effect of the combined therapy of metformin together with the SGLT-2 inhibitor AVE2268 after acute and subchronic intervention were evaluated in diabetes mouse models. Both interventions led to ameliorated glycaemia when compared to both mono therapies. The underlying causalities were identified using omics technologies of plasma and tissues. Further, a novel Ins2 mutation was characterized and the dependency of diabetes on carbohydrates was illustrated in TallyHo males.
