An early event in the pathology of the Alzheimer´s disease is the cleavage of amyloid precursor protein (APP) by ß-site APP-cleaving enzyme 1 (BACE1) and initiation of toxic amyloid-ß production. To investigate biology of this important enzyme, this study uses a newly developed proteomic method called Surface-Spanning Protein Enrichment with Click Sugars (SUSPECS) for surface glycoprotein enrichment to monitor changes of the cell surface neuronal proteome upon BACE1 inhibition. Furthermore, BACE1´s main substrate Seizure 6-like protein (SEZ6L) was investigated applying behavioural and proteomic studies.
