Protein aggregates in neurodegeneration exert toxicity on cells via loss of native function and gain of toxic function. To reduce complexity and study common effects of protein aggregation, this work aims to elucidate the cellular roles of toxic gain of function. We studied artificial beta-protein aggregates in situ using correlative cryo-electron tomography on focused ion beam milled mammalian cells. Beta-protein aggregates impaired the lysosomal system in neurons, recapitulating features of disease.
