DCM is a common cause of heart failure with high morbidity and mortality. A standard diagnostic method detecting pathological anti-ß1AR autoantibodies is not available. In this thesis, an in vitro immunoassay supporting the diagnosis of anti-ß1AR autoantibody-induced DCM was developed, optimized and validated. To enable the native presentation of the conformational epitope, the ß1AR was overexpressed and reconstituted into nanodiscs. 435 DCM and 90 control samples were characterized, a differentiation between autoantibody-positive and -negative subjects was possible.
