The 26S proteasome plays an essential role in protein degradation in eukaryotic cells. Using cryo-electron microscopy, comparative modeling, molecular dynamics and classification, pseudo atomic models of this 2.5MDa complex in three different conformations were determined. The developed classification approach combines an image-based classification with hierarchical clustering of rigidly fitted models. Finally, a functional model for states of recognition, commitment and degradation of substrates was proposed.
