Platelet activation at sites of vascular injury prevents live-threatening blood loss and promotes wound healing. However, their misregulation during vascular diseases may cause life-threatening complications like myocardial infarction and stroke. The two sphingosinkinase isoforms, sphingosine kinase 1 and 2 catalyze the phosphorylation of sphingosine to the bioactive metabolite sphingosine-1-phosphate. In a mouse model of sphingosine kinase 1/2-deficiency it was shown that these enzymes play an important role in the regulation of arterial thrombosis.
