Antibodies are essential effectors of the immune system. Within this study different isolated antibody domains were investigated in vitro. First, the role of conserved residues in the VH/VL interface concerning folding, stability and VH/VL binding was assessed. For the CH2 domain, a stability increase was generated by N- or C-terminal amino acid additions. The unfolded nature of the CH1 domain in isolation was studied by amino acid exchanges with CL. Here, a mutant with strong acceleration of the CL induced folding was identified. The obtained data is useful for antibody engineering.
