Particles of wear debris have been associated with loosening of implants and osteolysis. A new method for isolation of titanium alloy particles out of periprosthetic tissue retrieved from patients during revision surgery in case of failed total hip prosthesises was developed. The tissues were exposed to hydrochloric acid solution that degraded and solubilized them, liberating metal reaction products and leaving behind metal wear particles, which were seperated by centrifugation. Afterwards the particles were analyzed by raster electron microscopy and atomic absorption spectrophotometry. The influence of titanium particles on the human osteoblast like cell line SAOS-2 was studied. Dose dependent reactions on cell proliferation and cell metabolism were shown with biochemical methods like the colorimetric MTT method, the LDH cytotoxicity assay and the trypan blue exclusion assay.
