Intestinal stem cells (ISCs) maintain gut homeostasis and give rise to all intestinal epithelial cell types including hormone-secreting enteroendocrine (EE) cells. Diet-induced ISC dysfunction and EE disorder are implicated in the pathogenesis of the metabolic syndrome, which includes type 2 diabetes, and cancer, however the underlying mechanisms are not clear. This work reveals that high-fat diet in mice disturbs ISC homeostasis and EE lineage recruitment and identifies molecular mechanisms underlying this imbalance in obesity and pre-diabetes.
