Diese Thesis befasst sich mit der Untersuchung der Tumor-suppressor p53 Gene und der Bindung von p53 Protein an spezifische DNA Sequenzen. Wir stellen ein neuartiges elektrooptisches Oberflächenbiosensorsystem zur Detektion von Nukleinsäuresequenzen und Analyse von Protein-DNA-Interaktionen vor. Die Technik ermöglicht labelfreie Target-Probe Affinitätsmessungen in Echtzeit sowie die Größenbestimmung von gebundenen Molekülen auf einem Chip (die switchSENSE Technik).

This thesis reports on the detection of the tumor suppressor p53 gene and the DNA binding of p53 protein on electrically manipulated DNA interfaces. We introduce a novel electrooptical surface biosensor scheme for detecting nucleic acid sequences and analyzing protein-DNA interaction. The technique provides label-free target-probe affinity measurements in real-time and the size analysis of bound molecules on a chip (the switchSENSE technique).
