The goal of the thesis was, to find and characterise possible oncogenic alterations in genes of tyrosine kinases. The genetic alterations, which are making proto-oncogenes to oncogenes, may be used as targets in the development of novel cancer therapies. Therefore a large scale sequencing screen was set up, where all the 90 tyrosine kinase genes were analysed in cDNAs of 250 cancer cell lines. In one part of the thesis, the functional characterisation of two transforming mutations in the FES (feline sarcoma oncogene) tyrosine kinase is described. Further a mutation in the fibroblast growth factor receptor 4 (FGFR4) gene, which was discovered in the “Singapore Oncogenome Project” in the breast cancer cell line MDA-MB453, was confirmed to be the genetic reason of the corresponding breast cancer tumour.
