In this study a 16S rRNA oligonucleotide microarray for the differentiation of members of the phylum Acidobacteria was constructed. The microrarray results were verified by fluorescence in situ hybridisations as well as clone librariers. The evaluation and application of the microarray was carried out with a novel evanescent field DNA biosensor prototype, allowing the observation of hybridisation events in real time. Various soil samples , differing in soil types, plant diversity and contamination with 2,4,6-trinitrotoluene, were analysed.
