Abstract:
Ziel des von der DFG geförderten Projektes mit dem Titel Bestimmung polarer Pflanzenschutzmittel am Beispiel der Sulfonylharnstoffherbizide mit Immunextraktion-Tandem-Massenspektrometrie (IAC-LC-MS/MS) war es, klassenspezifische Antikörper herzustellen und als selektive Rezeptoren für die Anreicherung dieser Verbindungen aus Umwelt- und Lebensmittelproben zu verwenden. Dafür wurden zunächst drei verschiedene Immunogene synthetisiert und zur polyklonalen und monoklonalen Antikörperproduktion eingesetzt. Der generierte polyklonale anti-Sulfonylharnstoff-Antikörper mit der breitesten Erkennung wurde zur Entwicklung eines klassenspezifischen Screening-Immunoassays (ELISA) und eines Immunadsorbers auf Sol-Gel-Glas-Basis eingesetzt. Die Anwendbarkeit der Immunadsorber für die Off- und On-line-Anreicherung von Sulfonylharnstoffen aus Wasser und Lebensmittelextrakten wurde gezeigt. Zur Bestimmung wurden flüssigchromatographische Verfahren mit UV/DAD- und MS/MS-Detektion eingesetzt.

The thesis was based on a project financed by the DFG with the title Determination of Polar Pesticides by Immunoextraction-Tandem Mass Spectrometry (IAC-LC-MS/MS) Using Sulfonylurea Herbicides as Model Compounds. Its aim was the development of class-specific antibodies and their application as selective receptors for the enrichment of sulfonylureas from environmental and food samples. First of all three different immunogens were synthesized and employed for polyclonal and monoclonal antibody production. The gained polyclonal anti-sulfonylurea-antibody possessing the broadest specificity was used to establish a class-specific ELISA for rapid and cost-effective sample monitoring. For immunosorbent preparation corresponding antibodies were immobilized in sol-gel glasses. Advantages and limitations of immunosorbents in off- and on-line-applications were shown for water and food matrices spiked with sulfonylureas. Liquid chromatographic techniques combined with UV/DAD- and MS/MS-detection were used for the sulfonylurea determination.
