In der vorliegenden Arbeit wurde erstmals eine Apparatur etabliert, in der alveoläre Epithelzellen in-vitro mit luftgetragenen ultrafeinen Partikeln unter kontinuierlicher dosimetrischer Kontrolle exponiert werden konnten. Die Zellen wurden in einer Monolage an der Luft-Medium-Grenzschicht kultiviert und ihre Viabilität nach Exposition kontrolliert (WST1, Trypanblau). Die anschließend angewendete cDNA-Array-Technologie erlaubte ein Screening der Genexpressionen von exponierten Zellen. Mittels LC-PCR konnten signifikante dosisabhängige Effekte auf die Transkription der Stressproteine HSPA6 und HO1 durch die Exposition mit Partikeln nachgewiesen werden. Bei Expositionen der Zellen an der Luft-Medium-Grenzschicht genügte im Vergleich zu submersen Expositionen eine ca. 15-fach niedrigere Dosis um vergleichbare Effekte zu erhalten.

In this study a new in-vitro system was established to expose alveolar epithelial cells with airborne ultrafine particles under online dosimetric control. The cells were cultured in a monolayer at the air liquid interface and after exposure their viability was controlled (WST1, trypan blue). By applying cDNA-array technology cells exposed to airborne particles were screened for gene expression changes. Significant dose-dependent effects on the transcription of the stress proteins HSPA6 and HO1 were shown by LC-PCR. Compared with submersed exposure, at the air liquid interface a 15-fold lower dose was sufficient to obtain similar effects.
