Application of phase plate technology for transmission electron microscopy has been proposed for more than 80 years. However, it wasn’t until recently that this goal was realized at MPI of Biochemistry with the so-called Volta Phase Plate (VPP). Present thesis demonstrates application of the VPP. This entails achieving smallest protein structure solved to date as well as other biologically novel structures such as several active GPCR complexes. Furthermore, VPP was also applied to cryo-electron tomography and subtomogram averaging. In summary, advantages of the VPP for three-dimensional cryo-EM are discussed.
