Kurzfassung:
Die aktivitätsorientierte Fraktionierung von Kaffeegetränken und Modellsystemen führte zur Identifizierung von insgesamt 22 Bitterstoffen, die in Kaffeegetränken nachgewiesen werden konnten. Nach humansensorischer Bestimmung der individuellen Bitterschwellenwerte und Quantifizierung in Kaffeeaufgüssen wurde ihr Geschmacksbeitrag zur Bitterkeit von Kaffeeaufgüssen auf Basis von Dosis/Wirkungs-Überlegungen bewertet. Insgesamt 14 dieser Substanzen wurden im Rahmen dieser Arbeit erstmals als Bitterstoffe identifiziert. Desweiteren wurde der Einfluss der Prozessparameter bei der Röstung und Heißwasserextraktion von Kaffee sowie von Proteinen und proteinhaltigen Additiven als Rezepturbestandteil von Kaffeegetränken auf das Bitterstoffprofil und die dadurch induzierten Änderungen der wahrgenommenen Bitterkeit von Kaffeegetränken quantitativ und sensorisch erfasst. Zur quantitativen Untersuchung von Protein-Bitterstoff-Interaktionen in der komplexen Kaffeematrix wurde ein neuer Bioassay auf Basis von funktionalisierten magnetischen Partikeln entwickelt.

Activity-guided fractionation of coffee beverages and model systems led to the identification of 22 bitter tasting compounds which could be detected in coffee beverages. After sensory determination of the individual bitter thresholds and quantification in coffee brews the impact of these compounds on the bitter taste of coffee brews was estimated by means of dose/activity considerations. A total of 14 substances were reported for the first time as bitter compounds. Furthermore the influence of the process parameters during roasting and hot water extraction of coffee as well as of proteins and protein-containing additives as ingredients of coffee beverages on the profile of the bitter compounds and the perceived changes of the bitter taste of coffee beverages was quantitatively and sensorially analyzed. For quantitative investigation of interactions between proteins and bitter compounds in the complex coffee matrix a novel bioassay was developed based on functionalized magnetic particles.
