Abstract:
Die FLT3-ITD+ AML ist mit einer besonders schlechten Prognose assoziiert. Wir entdeckten das Chaperon pS305-SGTA, welches signifikant mehr in FLT3-ITD+ MV4-11- und in primären FLT3-ITD+ AML-Zellen phosphoryliert ist, wenn diese mit Sorafenib behandelt werden und auf EL08-1D2 Stromazellen kokultiviert werden. Die Phosphorylierung dieses Proteins könnte somit zur Resistenz der FLT3-ITD+ leukämischen Stammzellen (LSCs) beitragen. Demnach bietet pS305-SGTA einen interessanten Ansatz für die definitive Eradikation der FLT3-ITD+ LSCs.

We identified the chaperone pS305-SGTA whose phosphorylation was significantly higher in FLT3-ITD+ MV4-11 and in primary FLT3-ITD+ AML cells when treated with Sorafenib and co-cultured on EL08-1D2 stroma cells, indicating that phosphorylation of this protein might contribute to resistance of FLT3-ITD+ leukemic stem cells (LSCs). This strongly suggests pS305-SGTA as a potential target for definitive eradication of FLT3-ITD+ LSCs.
