This work aims to characterize patients with genetic variants in autoinflammation-related genes and the long-term effects of natalizumab therapy. Patients with mutations in autoinflammation-related genes display altered immunophenotypes and stronger IL-1 beta secretion upon pyrin inflammasome stimulation in case of MEFV variants. Natalizumab led to a decrease in serum IgM and intrathecal IgG production, while OCB and MRZ-reaction persisted. This points to an effect of the drug on plasmablasts, but not on plasma cells.

Patienten mit autoinflammationsassoziierten Varianten sollen besser charakterisiert und die Auswirkungen einer Langzeittherapie mit Natalizumab untersucht werden. Der Immunphänotyp von Patienten mit autoinflammationsassoziierten Varianten wies Veränderungen auf und im Fall von Patienten mit MEFV-Varianten zeigte sich eine erhöhte IL-1-beta-Ausschüttung nach Stimulation des Pyrin-Inflammasoms. Natalizumab führte zu einer Reduktion des IgM im Serum und der intrathekalen Synthese von IgG, was auf einen Effekt des Medikaments auf Plasmablasten, aber nicht auf Plasmazellen hinweist.
