Abstract:
This study aimed at deciphering, whether RNF43 has a tumor suppressive function in the stomach and by which signaling pathway it acts as a tumor suppressor. We found that RNF43 has a tumor suppressive function in the stomach and negatively regulates stemness in gastric cells. Furthermore, we showed that RNF43 is involved in DNA damage response and regulates susceptibility to DNA damage-inducing radio- and chemotherapy. Thus, we propose RN43 as a new potential biomarker to stratify gastric cancer patients for therapy selection.

Diese Studie zielte darauf ab, zu entschlüsseln, ob RNF43 im Magen als Tumorsuppressor fungiert und über welchen Signalweg es als solcher wirkt. Wir fanden heraus, dass RNF43 eine tumorunterdrückende Funktion im Magen hat und Stammzelleigenschaften in den Magenzellen negativ reguliert. Darüber hinaus zeigten wir, dass RNF43 an der DNA Schadensantwort beteiligt ist und die Anfälligkeit für DNA-Schäden verursachende Radio- und Chemotherapie reguliert. Daher schlagen wir RN43 als neuen potenziellen Biomarker vor, der Magenkrebspatienten für die Therapiewahl stratifiziert.
