Abstract:
The nuclear lamin proteins are modified by a series of posttranslational modifications. The mutant protein progerin is permanently associated with the nuclear envelope (NE), causing severe nuclear alterations. We analyzed the spatiotemporal distribution of the EGFP fusion lamin A peptides to obtain a better understanding of the abnormal interaction and retention of progerin to the NE. The farnesylated C-terminal lamin A peptides induce mis-shaped nuclei and induce nucleophagic degradation of the toxic proteins.

Die nuklearen Lamin Proteine werden mit verschiedenen Gruppen posttranslational modifiziert. Das mutierte Protein Progerin ist permanent an die nukleare Membran (NM) gebunden, was schwerwiegende Veränderungen des Zellkerns bewirkt. Wir haben die raumzeitliche Verteilung von Lamin A EGFP Fusionspeptiden analysiert, um ein besseres Verständnis für die abnormale Interaktion und permanente Bindung von Progerin an die NM zu gewinnen. Die farnesylierten C-terminalen Lamin A Peptide führen zu einer Verformung des Zellkerns und induzieren Nukleophagischen Abbau von toxischen Proteinen.
