Abstract:
Die in der akuten Hepatitis B zur Ausheilung führende starke T-Zell-Antwort ist in chronisch Hepatitis-B-Infizierten nicht vorzufinden. Ein adoptiver T-Zell-Transfer von HBV-spezifischen T-Zellen könnte daher im Gegensatz zu der aktuellen medikamentösen Therapie eine kurative Therapie darstellen. Es wurden 11 HBV-spezifische T-Zell-Rezeptoren gegen insgesamt 3 immunogene HBV-Peptide identifiziert und für den adoptiven T-Zell-Transfer optimiert, die transduziert in T-Zellen diese in hoch avide polyfunktionale zytotoxische HBV-spezifische Effektorzellen umwandelten und somit potentiell geeignet Antigenrezeptoren für eine adoptive T-Zell-Therapien der Hepatitis B sind.

The strong T cell response leading to cure in acute hepatitis B is not detected in chronically hepatitis-B-infected individuals. Adoptive T-cell transfer of HBV-specific T cells could therefore represent a curative therapy in contrast to current drug therapy. Eleven HBV-specific T-cell receptors against a total of 3 immunogenic HBV peptides were identified and optimized for adoptive T-cell transfer, which when transduced into T cells converted them into highly avid polyfunctional HBV-specific effector cells and thus represent suitable antigen receptors for adoptive T-cell therapy of HBV.
