Since cancer T-cell immunotherapy revealed promising clinical outcomes, non-invasive monitoring technologies are required for a better understanding of the efficacy and intravital behavior of the transferred TCR-transgenic T cells in vivo. A method has been developed to track engineered human T cells in vivo at the targeted tumor site by PET using the Zirconium-89 radiolabeled anti-TCRmu-F(ab’)2. Characterization studies have been performed regarding tracer quality as well as sensitivity, image correlation and accuracy to ex vivo quantified T cells. 89Zr-Df-aTCRmu-F(ab’)2 showed altogether suitable properties for in vivo monitoring of the engineered T cells speaking for the potential of clinical translation of the technology.

Da die T-Zell-Immuntherapie bei Krebserkrankungen vielversprechende klinische Ergebnisse zeigte, sind nicht-invasive Bildgebungstechnologien für das verbesserte Verständnis der Wirksamkeit und dem intravitalen Verhalten der transferierten TZR-transgenen T-Zellen erforderlich. Es wurde eine Methode entwickelt für das in vivo Tracking der humanen transgenen T-Zellen im PET unter Verwendung von Zirkonium-89 radioaktiv-markiertem anti-TCRmu-F(ab’)2. Es wurden Charakterisierungsstudien zur Tracer-Qualität sowie zur Sensitivität, Bild-Korrelation und Genauigkeit gegenüber ex vivo quantifizierter T-Zellen durchgeführt. 89Zr-Df-aTCRmu-F(ab’)2 zeigte insgesamt gute Eigenschaften für das in vivo Monitoring der transgenen T-Zellen, was für das Potenzial der klinischen Translation der Anwendung spricht.
