Alzheimer’s disease is the most common form of neurodegenerative dementia and proteolytic processing of the amyloid precursor protein (APP) is a driving process of the molecular pathological mechanisms. ADAM10, BACE1, and MT5-MMP are three APP-cleaving proteases and represent potential drug targets to modulate the disease. Their substrates and functions are still not fully known, which poses a risk for side effects. This dissertation aims to identify and/or validate additional substrates of the three proteases in order to elucidate their functions and allow safe disease modulation.

ADAM10, BACE1 und MT5-MMP sind drei APP-spaltende Proteasen und sind potentielle Medikamentenziele, um die Krankheit zu modulieren. Ihre Substrate und Funktionen sind noch nicht vollständig bekannt, was ein Risiko für Nebenwirkungen darstellt. Ziel dieser Dissertation ist es, weitere Substrate der drei Proteasen zu identifizieren und/oder zu validieren, um deren Funktionen aufzuklären und eine sichere Modulation der Krankheit zu ermöglichen.
